Is there anything in modern-day football regulations about a minimum number of senior players being required before a league game can take place? Or could Derby be forced to draft in U18s to bulk up their squad if nobody else is available?

they are in negotiations with HMRC and no doubt will be allowed to sign enough players to have a squad.

No they have sold Lucas Paqueta, Gabriel Barbara and Leo Duarte over recent seasons which were installment outgoings so they are in no rush to sell.

Bettinelli is off to Chelsea! Seems he's swapping doing nothing in the Middlesbrough goal to doing nothing in the stands for 90% of games as 3rd choice behind Kepa and Mendy.

They signed Barbosa back for 18m to be fair. Hes up top for them this season

I was reading up on this the other day, once a player makes an appearance for the first team they are classed as a senior player. They played a load of the kids in cup games last season so technically have about 17/18 senior players. I'm sure i read that a squad is expected to have a minimum of 15 senior players but I could be misremembering that.
